چکیده

Plasma fibronectin binds to gelatin and is thought to mediate the removal of blood-borne particles by the reticuloendothelial system. This removal has primarily been studied using gelatin-coated colloidal particles. Fibronectin also binds to gram-positive bacteria. We tested the possibility that fibronectin might act as an opsonin for phagocytosis of bacteria by neutrophils. When fibronectin in physiologic concentrations was present in reaction mixtures containing Staphylococcus aureus and human neutrophils, staphylococci adhered to. but were not ingested by, neutrophils. Chemiluminescence of . mixtures of neutrophils and S. aureus was enhanced by fibronectin. Although S. aureus were not killed by attachment, the number of colony-forming units fell as the bacteria were concentrated on the neutrophil surfaces. Washing of the neutrophils did not remove attached staphylococci. Sonication, however,

Construction of a New Fusion Protein Vector Associated to Fibronectin Binding Protein A and Clumping Factor A Derived from Staphylococcus aureus NCTC8325

Objective(s) Staphylococcus aureus is a leading cause of many nosocomial and community acquired infections. According to many reports, antibiotic therapy can not guarantee the eradication of S. aureus infections. Thus designing an adhesin based vaccine could restrain the S. aureus infections. This study designed for construction of a new fusion protein vaccine against S. aureus infections base...

Peptide analogs to a fibronectin receptor inhibit attachment of Staphylococcus aureus to fibronectin-containing substrates.

Binding of cells of Staphylococcus aureus to fibronectin has been proposed as a mechanism of bacterial adhesion to host tissues. In this study, we have attempted to define the role of a recently identified fibronectin receptor in the adhesion of staphylococcal cells to fibronectin-containing substrates by using different receptor analogs as potential inhibitors of bacterial adherence. The resul...

Growth and adherence of Staphylococcus aureus were enhanced through the PGE2 produced by the activated COX-2/PGE2 pathway of infected oral epithelial cells

Staphylococcus aureus is a major pathogen of varieties of oral mucous infection. Prostaglandin E2 (PGE2) is a pro-inflammatory factor and Cyclooxygenase 2 (COX-2) is a critical enzyme of PGE2 biosynthesis. The purpose of this study is to investigate whether Staphylococcus aureus can increase PGE2 production of oral epithelial cells and how PGE2 functions in the growth and adherence of Staphyloc...

Staphylococcus aureus Fibronectin-Binding Protein A Mediates Cell-Cell Adhesion through Low-Affinity Homophilic Bonds

UNLABELLED Staphylococcus aureus is an important opportunistic pathogen which is a leading cause of biofilm-associated infections on indwelling medical devices. The cell surface-located fibronectin-binding protein A (FnBPA) plays an important role in the accumulation phase of biofilm formation by methicillin-resistant S. aureus (MRSA), but the underlying molecular interactions are not yet estab...

Cellular invasion by Staphylococcus aureus reveals a functional link between focal adhesion kinase and cortactin in integrin-mediated internalisation.

Nosocomial infections by Staphylococcus aureus, a Gram-positive pathogen colonising human skin and mucosal surfaces, are an increasing health care problem. Clinical isolates almost invariably express fibronectin-binding proteins that, by indirectly linking the bacteria with host integrin alpha5beta1, can promote uptake of the microorganisms by eukaryotic cells. Integrin engagement by pathogenic...
